<Adaptive Immunity>

- activated only after exposure to specific pathogen

- antigen: adaptive immnune에서 pathogen 역할을 하는 molecule

- active immunity: 면역계가 스스로 antibody 생성

passive immunity: 항체를 주입하는 것. e.g. 모유 속 항체, 항체 치료

- humoral / cell-mediated로 나뉨

- innate immnuity와 비교: slower, recognize specific pathogen, memory cell

- herd immunity: 일정 비율 이상 접종 완료되면, 더이상 질병이 퍼지지 않는 것

 

* lymphatic system

- 구성

① lymphatic vessel: valve 존재

② lymph node: macrophage, lymphocyte가 뭉쳐있는 둥근 기관

③ bone marrow: leukocyte 생성

④ organ: adenoid, tonsil, thymus, spleen, appendix

⑤ lymph: interstitial fluid와 유사. 산소와 nutrient 함량이 낮음

- function

① blood vessel에서 온 fluid를 다시 circulatory system으로

② immune

 

* lymphocyte

bone marrow의 stem cell에서 lymphocyte 생성

T cell: thymus에서 성숙 → lymph node에서 pathogen 만나면 최종 성숙 → cell-mediated immune response

B cell: bone marrow에서 성숙 → lymph node에서 pathogen 만나면 최종 성숙 → humoral imune response

 

* B cell

- antigen의 epitope이 antibody의 antigen-binding site에서 결합해 항원 인식

- antibody는 pathogen을 죽이는 것이 아니라, 없애는 것을 도움

중화, 응집으로 phagocytosis 유도, complement system을 활성화시켜 cell lysis 유도

 

* T cell

* APC(antigen-presenting cell)

- 표면에 MHC molecule에 antigen을 결합해 helpler T cell에 제시함

- MHC는 사람마다 다르기 때문에 immune tolerance 일어남

- helper T cell과 결합할 때 antigen, MHC 각각 결합해 double recognition

① helper T cell

- APC와 결합함으로 인해 

1) 자기 자신 활성화(clonal selection)

2) B cell의 antibody 분비량 증가

3) cell-mediated immune response 유도

→ humoral, cell-mediated immune response 모두 관여

② cytotoxic T cell

infected cell과 결합해 활성화 → perforin(막에 구멍 뚫음), enzyme 등을 분비해 infected cell 죽임

(cell-mediated immune response만)

 

 

* clonal selection

① 여러 cell 중 antigen와 결합 가능한 receptor를 가진 cell 선택

② 선택된 cell을 빠르게 분열시켜 양을 늘림

③ 일부는 effector cell로 분화해 antibody 생성 → 이후 macrophage에 의해 먹힘

일부는 memory cell로 분화해 lymph node에서 보관됨

- memory cell로 인해 secondary immune response가 primary immune response보다 faster, stronger, more proloned

 

* HIV

- human immunodeficiency virus의 약자

- AIDS(acquired immunodeficiency syndrome)을 일으킴

- helper T cell을 공격해 모든 adaptive immunity를 무너뜨림

- opportunistic infections(면역력이 낮아진 사람만 감염 증상을 보이는 것)이 나타남

- 치료가 어려운 이유

1) RNA virus이기 때문에 mutation이 빠르게 일어남

2) editing function이 없기 때문에 mutation 비율이 높음

- 치료 사례

1) AIDS 치료 중 암에 걸려 골수이식 → AIDS에 내성이 있는 사람의 골수를 이식 받아 helper T cell 다시 형성

2) 유전자 교정
